Prelude Therapeutics announced on October 13 a clinical trial collaboration and supply agreement with Eli Lilly, according to the company’s announcement.

The deal

Lilly will supply its CDK4/6 inhibitor abemaciclib, sold as Verzenio, and its oral estrogen receptor degrader imlunestrant, sold as Inluriyo, for a Phase 1 trial combining each with Prelude’s PRT13722, an oral degrader of the KAT6A protein. Prelude retains full global development and commercial rights to PRT13722. No payments were disclosed.

The trial

The FDA has cleared the study under Prelude’s investigational new drug application. Enrollment is expected to begin in the fourth quarter of 2026 with single-agent dose escalation, followed by combination cohorts in patients with hormone receptor-positive, HER2-negative breast cancer. The company cites preclinical work showing activity alone and in combination with both drug classes; there are no human data on efficacy or safety yet.

What to compare next

For readers following PRLD, a supply agreement with a large pharmaceutical company validates the trial design but is not a licensing deal, and the company’s cash runway remains the first question for a small biotech entering a multi-arm study. For LLY, supplying drugs to a partner’s trial is routine and immaterial to results; the interest is whether a new combination extends the useful life of its breast cancer franchise.
